The HIV polytope vaccine is an experimental recombinant viral vector vaccine designed to elicit a broad CD8+ cytotoxic T-lymphocyte (CTL) response against HIV-1. Developed by researchers at the Queensland Institute of Medical Research and affiliated Australian centers, the vaccine utilizes a "polytope" approach, where multiple minimal HLA A2-restricted CTL epitopes—specifically seven in the rVV.HIV.pt construct—are conjoined into a single artificial protein. By targeting multiple epitopes simultaneously, the vaccine aims to provide protection against diverse HIV challenge isolates and prevent the emergence of CTL escape mutants. Preclinical testing in HLA A2-transgenic mice has demonstrated the vaccine's ability to induce multi-specific cellular immunity, validating the construct's design for potential use in the prevention and treatment of HIV-1 infection and AIDS.
