HIV-specific chimeric antigen receptor (CAR) T cells are an autologous cell therapy developed by the Shanghai Public Health Clinical Center (SPHCC) for the treatment of HIV infection and AIDS-related B-cell lymphoma. These T cells are genetically engineered to express a chimeric antigen receptor that targets the HIV-1 envelope glycoprotein gp120. A prominent construct used by SPHCC, known as VC-CAR-T, utilizes the V1 and V2 domains of the human CD4 molecule as the antigen-binding domain, enabling the T cells to selectively recognize and eliminate HIV-infected cells. This approach aims to reduce the latent viral reservoir and potentially achieve a functional cure for HIV. In the context of AIDS-related B-cell lymphoma, these CAR-T cells are investigated for their ability to clear HIV-infected cells and potentially improve outcomes in patients with HIV-associated malignancies.
