The HIVIS DNA HIV-1 vaccine is a multigene, multiclade plasmid DNA-based vaccine developed by the Swedish Institute for Infectious Disease Control (SMI) in collaboration with Karolinska Institutet and Karolinska University Hospital. It consists of seven plasmids encoding multiple HIV-1 antigens, including envelope gp160 of subtypes A, B, and C; Rev B; Gag A and B; and a mutated reverse transcriptase (RTmut) B. The vaccine is designed to stimulate endogenous production of these HIV-1 proteins to elicit broad cellular and humoral immune responses. In clinical trials, such as the HIVIS 01/02 Phase I study, the vaccine was evaluated as a prime in a DNA-prime/MVA-boost strategy, often incorporating recombinant GM-CSF as an adjuvant to enhance immunogenicity. It is intended for prophylactic use against HIV-1 infection and AIDS.
