The HIVIS-DNA vaccine is an investigational multiclade, multigene DNA-based vaccine designed for the prevention and treatment of HIV-1 infection. Developed by the Swedish Institute for Infectious Disease Control (SMI) in collaboration with the Karolinska Institutet and Karolinska University Hospital, the vaccine consists of seven plasmid DNA constructs encoding multiple HIV-1 antigens. These include envelope glycoproteins (gp160) from subtypes A, B, and C; Rev (subtype B); Gag (subtypes A and B); and a mutated reverse transcriptase (RTmut, subtype B). The vaccine is intended to induce broad cellular and humoral immune responses across multiple HIV-1 clades. It is typically administered as a priming agent in a heterologous prime-boost regimen, often followed by a Modified Vaccinia Ankara (MVA-CMDR) boost. Clinical studies have evaluated its safety and immunogenicity in healthy volunteers and children with vertically acquired HIV infection receiving antiretroviral therapy.
