hJAA-F11 is a humanized monoclonal antibody developed by For-Robin, Inc. that specifically targets the Thomsen-Friedenreich antigen (TF-Ag), an oncofetal carbohydrate antigen (Galβ1-3GalNAcα1-Ser/Thr) found on approximately 85% of human carcinomas but cryptic on normal tissue. Originally developed as a mouse monoclonal antibody at the University at Buffalo, it has been humanized into constructs such as hJAA-F11-H2aL2a and hJAA-F11-H3L3 to reduce immunogenicity while maintaining high affinity for the alpha-linked TF-Ag. The antibody's mechanism of action involves direct binding to tumor cells and the recruitment of immune effectors through antibody-dependent cellular cytotoxicity (ADCC). Additionally, hJAA-F11 is internalized by cancer cells, which facilitates its potential use in antibody-drug conjugate (ADC) applications. Preclinical studies have demonstrated that the antibody significantly decreases tumor growth and metastasis in xenograft models of breast and lung cancer.
