Overview

HL-400 is an orally bioavailable small molecule inhibitor of the NLRP3 (NACHT, LRR, and PYD domains-containing protein 3) inflammasome. Developed by Hangzhou Highlightll Pharmaceutical, HL-400 is designed to prevent the assembly and activation of the NLRP3 inflammasome complex, a critical component of the innate immune system. The NLRP3 inflammasome triggers the activation of caspase-1, which in turn leads to the maturation and secretion of potent pro-inflammatory cytokines, specifically interleukin-1β (IL-1β) and interleukin-18 (IL-18), and initiates pyroptosis. By blocking this pathway, HL-400 aims to mitigate chronic inflammation associated with a wide range of conditions, including metabolic, cardiovascular, and neurodegenerative diseases. The compound is currently in early-stage clinical development, with Phase 1 studies evaluating its safety, tolerability, and pharmacokinetics in healthy volunteers.
