HL140 is a fixed-dose combination (FDC) drug developed by Hanlim Pharm. Co., Ltd. for the treatment of primary hypercholesterolemia. It combines two lipid-lowering agents with complementary mechanisms of action: rosuvastatin and ezetimibe. Rosuvastatin is an HMG-CoA reductase inhibitor (statin) that works by inhibiting the rate-limiting enzyme in hepatic cholesterol synthesis, which leads to an increase in LDL receptor expression on hepatocytes and enhanced clearance of LDL-C from the circulation. Ezetimibe is a selective cholesterol absorption inhibitor that targets the Niemann-Pick C1-Like 1 (NPC1L1) protein in the small intestine, thereby reducing the delivery of intestinal cholesterol to the liver. The dual inhibition of cholesterol synthesis and absorption provided by HL140 results in a more significant reduction in LDL-C levels compared to monotherapy with either component. HL140 has been evaluated in Phase 3 clinical trials to demonstrate its efficacy and safety in patients with primary hypercholesterolemia.
