Overview

**HL237 + tacrolimus** is a combination treatment comprising HL237, a small molecule STAT3 inhibitor being developed for autoimmune and inflammatory diseases (most notably rheumatoid arthritis), and tacrolimus, a calcineurin inhibitor widely used as an immunosuppressant primarily to prevent organ transplant rejection. HL237 acts by inhibiting Signal transducer and activator of transcription 3 (STAT3), modulating immune cell differentiation (Th17 suppression and Treg activation), and thereby exerting effects against autoimmune activity. Tacrolimus binds to FK506-binding proteins (FKBP), inhibiting calcineurin phosphatase, blocking T-cell activation, proliferation, and related cytokine gene transcription (especially IL-2). The combination is being studied to evaluate pharmacokinetic interactions and potential synergistic immunomodulatory or anti-inflammatory effects, especially for autoimmune conditions such as rheumatoid arthritis[1][2][4][6][8][9].
