HL2541 is a monoclonal antibody directed against the IgC domain of CD33, specifically designed to recognize the CD33-D2 splice variant isoform. CD33-D2 is a shorter isoform of CD33 that lacks the IgV domain (the binding site for gemtuzumab ozogamicin) due to a splicing polymorphism (rs12459419). HL2541 was developed by researchers at the University of Florida to address the lack of clinical benefit from standard CD33-targeted therapies in patients expressing this variant. The antibody has demonstrated the ability to internalize upon binding to CD33-D2 in acute myeloid leukemia (AML) cell lines and has shown in vivo targeting potential in mouse models. It is being explored for the development of novel antibody-drug conjugates (ADCs), T-cell engagers, and immunoPET imaging modalities for the treatment and monitoring of AML.
