Drug intelligence / Profile preview
HLA-A2-Ig fusion protein (often written HLA-A2:Ig) is a recombinant soluble dimeric fusion protein. It is widely utilized as a research-use-only (RUO) reagent, commercialized by BD Biosciences under the brand name BD DimerX, to detect, stain, and stimulate antigen-specific CD8+ T cells by binding to the T-cell receptor (TCR). The reagent consists of three extracellular MHC class I HLA-A2 domains fused to the VH regions of mouse IgG1, supplemented with recombinant β2-microglobulin (β2M) to facilitate peptide binding. Additionally, academic researchers at CHU Nice and CNRS have developed cytotoxic HLA-A2-Ig fusion protein variants (HLA-A2-Ig dimers) as potential therapeutic candidates for desensitization in highly sensitized transplant patients, designed to selectively target and deplete anti-HLA-A2 memory B cells and antibody-producing hybridomas via their B-cell receptors (BCR).
