Overview

HLA-A2 restricted glioma antigen peptides vaccine is an investigational cancer immunotherapy designed to stimulate the immune system to recognize and attack tumor cells in patients with gliomas who express the human leukocyte antigen A*02 (HLA-A2) allele. The vaccine consists of synthetic peptide fragments derived from tumor-associated antigens commonly expressed in malignant brain tumors—such as EphA2, interleukin-13 receptor alpha 2 (IL13Rα2), and survivin—that are presented by the patient's own HLA-A*02 molecules on the surface of tumor cells. By vaccinating with these specific peptides, often combined with adjuvants like poly-ICLC or Montanide ISA-51 to enhance immune response, the therapy aims to induce a cytotoxic T lymphocyte response against tumor cells expressing these antigens. The approach has been studied primarily in pediatric and adult patients with newly diagnosed or recurrent high-grade or low-grade gliomas[5][6][7].
