HLA-matched multivirus-specific cytotoxic T lymphocytes (VSTs) is an investigational cell therapy developed by the Center for Cell and Gene Therapy at Baylor College of Medicine. The therapy consists of allogeneic, banked, off-the-shelf T cells derived from healthy donors and expanded ex vivo to target immunodominant antigens from multiple viruses, specifically Epstein-Barr virus (EBV), cytomegalovirus (CMV), adenovirus (Adv), and BK virus. These polyclonal cells, containing both CD4+ and CD8+ T lymphocytes, are adoptively transferred to immunocompromised patients, such as hematopoietic stem cell transplant recipients, to restore antiviral immunity and treat persistent or refractory viral infections. Clinical data from the TETRAVI program has shown high response rates (approximately 67-92%) without significant graft-versus-host disease, providing a therapeutic option for patients who have failed conventional antiviral drugs.
