HLA-pan-DP CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target HLA-DP, a class II major histocompatibility complex (MHC) protein expressed across a wide range of B-cell lineages and various hematological malignancies. Developed through a collaboration between the University of California San Francisco, Kanazawa University, and Wakunaga Pharmaceutical, the therapy utilizes a single-chain variable fragment (scFv) derived from the B7/21 monoclonal antibody clone. Preclinical studies have demonstrated significant cytotoxicity against multiple myeloma (MM), B-cell lymphomas, and blastic plasmacytoid dendritic cell neoplasm (BPDCN). Notably, HLA-DP expression is often significantly higher at relapse in myeloma patients compared to diagnosis. Because HLA-DP is also expressed on normal hematopoietic cells such as B-cells and dendritic cells, this therapy is primarily envisioned as a potent bridge to hematopoietic stem cell transplantation (HSCT).
