Drug intelligence / Profile preview
HLM1196 is a novel, highly potent small molecule anti-fibrotic compound belonging to the N-(2-butoxyphenyl)-3-(phenyl)acrylamide (N23P) class. Developed by researchers at Helmholtz Zentrum München (Helmholtz Munich) and partners, HLM1196 was optimized via medicinal chemistry from the parent scaffold of tranilast to enhance potency while eliminating NOX4 and pro-Wnt liabilities. Mechanistically, HLM1196 acts as a selective microtubule destabilizer. It induces cell rounding and collapses cellular tensegrity in primary human lung fibroblasts, leading to a mitotic catastrophe signature in CTHRC1+ myofibroblasts and reversing integrin-linked signaling in KRT17+ aberrant basaloid cells. HLM1196 is being investigated for the treatment of idiopathic pulmonary fibrosis (IPF) and progressive pulmonary fibrosis.
