HLS5 (Hematopoietic Lineage Switch 5), also known as TRIM35, is a tumor suppressor gene being developed as a gene therapy by Molecular Discovery Systems (MDSystems), an investment of BPH Energy (formerly BioPharmica). HLS5 is a member of the RBCC (Ring finger, B box, coiled-coil) family of genes located on chromosome 8p21, a region frequently implicated in various leukemias and solid tumors. The therapeutic approach involves restoring or enhancing the function of this tumor suppressor, which is often downregulated in various cancers, particularly liver cancer, breast cancer, and melanoma. Beyond oncology, research suggests HLS5 plays a role in multiple signaling pathways relevant to neurodegenerative diseases like Huntington's and Parkinson's, as well as the regulation of HIV replication. The program is currently in the preclinical development stage, with validation efforts conducted in collaboration with the Harry Perkins Institute of Medical Research.
