HLX109 is a monoclonal antibody (mAb) targeting interleukin-1 receptor accessory protein (IL-1R3, also known as IL1RAP), currently in development by Shanghai Henlius Biotech. IL-1R3 is a common coreceptor for several members of the interleukin-1 receptor family, including IL-1R1, IL-1R6 (IL-36R), and ST2 (IL-33R). By binding to IL-1R3, HLX109 inhibits the signaling of IL-1α, IL-1β, IL-33, and IL-36, which are critical drivers of chronic inflammation and tumor progression. Henlius acquired global exclusive rights to the asset from Mabwell Bioscience in 2022. The drug is positioned for potential applications in both oncology and inflammatory diseases, although specific lead indications have not been formally announced.
