Overview

HLX22 + trastuzumab deruxtecan is a combination therapy under investigation for the treatment of HER2-low, hormone receptor-positive, locally advanced or metastatic breast cancer. HLX22 is an innovative anti-HER2 monoclonal antibody that binds to HER2 extracellular subdomain IV at a different epitope from trastuzumab, enabling simultaneous HER2 dimer binding and promoting HER2 dimer internalization and degradation. Trastuzumab deruxtecan (T-DXd), an antibody-drug conjugate, consists of a humanized HER2-targeting monoclonal antibody linked to the cytotoxic topoisomerase I inhibitor deruxtecan. The combination aims to provide synergistic anti-tumor effects by leveraging distinct HER2 binding sites and enhanced cytotoxic payload delivery. The therapy is primarily developed by Shanghai Henlius Biotech and is currently in phase 2 clinical trials for breast cancer and approved for phase 2 trials in HER2-expressing solid tumors, expanding its investigational scope in oncology[1][4][7].
