HLX26 is a recombinant human monoclonal antibody that targets lymphocyte activation gene 3 (LAG-3, also known as CD223), an immune checkpoint receptor expressed on activated T cells and some NK cells. By binding to the extracellular domain of LAG-3, HLX26 blocks LAG-3-mediated signaling pathways and restores T-cell cytotoxic function. This mechanism enhances the immune system's ability to attack tumor cells. Preclinical studies have demonstrated anti-tumor activity and favorable safety profiles for HLX26 both as monotherapy and in combination with other immunotherapies such as serplulimab (an anti-PD-1 antibody). The drug is being developed primarily for solid tumors—including non-small cell lung cancer (NSCLC) and colorectal cancer—and lymphomas[2][3][4][5].
