HLX60 is a novel, self-developed humanized monoclonal antibody targeting glycoprotein A repetitions predominant (GARP; also known as leucine-rich repeat-containing protein 32 or LRRC32), which acts as a transforming growth factor beta (TGF-β) activator. GARP is highly expressed on activated regulatory T cells (Tregs) and platelets and functions by binding latent TGF-β1, facilitating its activation in the tumor microenvironment. HLX60 binds to GARP and specifically blocks the release of GARP-mediated TGF-β1, thereby relieving immunosuppression caused by TGF-β1, reversing the immunosuppressive tumor microenvironment (TME), and enhancing antitumor immune responses. Additionally, HLX60 induces antibody-dependent cell-mediated cytotoxicity (ADCC), enabling depletion of GARP-positive tumor cells and immunosuppressive cells such as Tregs. Preclinical studies have shown that HLX60 has antitumor effects with a good safety profile. It is being developed for solid tumors and lymphomas by Shanghai Henlius Biotech[1][2][4][5][7].
