HLX6018 is a novel, first-in-class monoclonal antibody developed by Shanghai Henlius Biotech that specifically targets the Glycoprotein A repetitions predominant (GARP)/Transforming growth factor beta-1 (TGF-β1) complex. By binding to this complex, HLX6018 blocks the release of active TGF-β1, a potent profibrotic cytokine. This mechanism inhibits fibroblast proliferation and the secretion of extracellular matrix (ECM) components, which are central to the progression of fibrotic diseases. HLX6018 is primarily being developed for the treatment of idiopathic pulmonary fibrosis (IPF) and has shown promising anti-fibrotic effects in non-clinical models of both pulmonary and renal fibrosis. As of 2024, it is undergoing Phase 1 clinical trials to evaluate safety, tolerability, and pharmacokinetics in healthy volunteers.
