Overview

HLX701 is a recombinant humanized anti-CD47 SIRPα-Fc fusion protein developed by Shanghai Henlius Biotech. It is designed to target CD47, a cell surface protein often overexpressed on cancer cells that acts as a "don't eat me" signal by interacting with SIRPα on macrophages to inhibit phagocytosis. By blocking the CD47-SIRPα axis, HLX701 enhances the ability of the innate immune system to identify and clear tumor cells. The fusion protein format, which utilizes the extracellular domain of SIRPα, is intended to provide a better safety profile—specifically by reducing the risk of hemagglutination and anemia—compared to first-generation anti-CD47 monoclonal antibodies. HLX701 is currently being evaluated in clinical trials for the treatment of advanced solid tumors and hematologic malignancies.
