**HM004-6599** is an enriched formulation of HMPL-004, developed to contain over 70% diterpenoids (versus ~15% in the original HMPL-004). Diterpenoids are plant-derived compounds with anti-inflammatory effects. HM004-6599 has demonstrated superior inhibition of NF-kB activation, reduced pro-inflammatory cytokine IL-1β production, and decreased TNF-α dependent chemokine production including CCL-20 in pre-clinical models. These mechanisms suggest utility in treating inflammatory and autoimmune conditions such as ulcerative colitis or Crohn’s disease. HM004-6599 is being developed by Hutchison China MediTech (HutchMed) and represents an optimization of the original botanical drug candidate for improved efficacy and lower dosing requirements[1][7][13][9].
