HM100714 is an orally bioavailable, selective, and covalent small molecule inhibitor of Human Epidermal Growth Factor Receptor 2 (HER2), currently in preclinical development by Hanmi Pharmaceutical. It is being investigated for the treatment of HER2-altered cancers, including those with central nervous system (CNS) metastases. Preclinical studies indicate that HM100714 has the potential to overcome resistance to existing therapies like trastuzumab deruxtecan (Enhertu) and exhibits significant blood-brain barrier penetration, addressing a critical need in patients with HER2-positive brain metastases.
