Overview

HM16390 is a long-acting interleukin-2 (IL-2) analog developed by Hanmi Pharmaceutical for the treatment of solid tumors. It is a recombinant human IL-2 (rhIL-2) variant chemically conjugated to the constant region of human immunoglobulin G4 (IgG4 Fc) via a flexible non-peptidyl linker. The drug is engineered to preferentially bind to the interleukin-2 receptor beta chain (IL-2Rβ/CD122), enhancing anti-tumor efficacy, while maintaining an optimized affinity for the interleukin-2 receptor alpha chain (IL-2Rα/CD25), which improves safety and reduces adverse events such as vascular leak syndrome and cytokine release syndrome. Preclinical studies have demonstrated that HM16390 elicits robust lymphocyte expansion, increases tumor-infiltrating immune cells, remodels the tumor microenvironment, and shows synergistic effects when combined with immune checkpoint inhibitors. It is administered as a once-per-treatment cycle subcutaneous injection using Hanmi’s LAPSCOVERY platform technology[1][5][6][7].
