HM30-tesirine is an antibody-drug conjugate (ADC) designed to target the xCT subunit (encoded by the SLC7A11 gene) of the system xc- cystine/glutamate antiporter. Developed by AgilVax, the drug consists of the HM30 monoclonal antibody conjugated to tesirine (SG3249), a potent pyrrolobenzodiazepine (PBD) dimer cytotoxin. The xCT transporter is a critical component of the cellular antioxidant defense system and is frequently upregulated in various malignancies, particularly non-small cell lung cancer (NSCLC) harboring NRF2-KEAP1 pathway mutations. By binding to xCT on the tumor cell surface, HM30-tesirine is internalized and releases its tesirine payload, which induces lethal DNA interstrand cross-links. Preclinical studies have demonstrated that HM30-tesirine effectively suppresses tumor growth and improves survival in aggressive, therapy-resistant models of NSCLC.
