hMaxi-K is a gene therapy consisting of a naked DNA plasmid encoding the human cDNA for the alpha (pore-forming) subunit of the human smooth muscle Maxi-K channel (also known as BK or hSlo channel). The therapy is designed to increase expression of this potassium channel in smooth muscle cells, leading to increased potassium efflux, decreased calcium influx, and subsequent relaxation of smooth muscle. This mechanism underlies its intended use for conditions such as erectile dysfunction (ED) and overactive bladder (OAB), where enhanced smooth muscle relaxation can improve symptoms. The product was developed by Ion Channel Innovations and later advanced by Sumitomo Pharma America (formerly Urovant Sciences). Clinical studies have shown promising safety results and preliminary efficacy in both ED and OAB; however, development has been discontinued for these indications[1][2][3][4][5][6].
