HMBD-001 is a humanized IgG1 monoclonal antibody developed by Hummingbird Bioscience using its Rational Antibody Discovery platform. It is engineered to bind strongly and specifically to the dimerization interface of HER3 (also known as ERBB-3), thereby interfering with HER3's ability to dimerize and fully blocking its activation regardless of NRG1 binding[1][2][3]. By targeting HER3, which is implicated in various solid tumors including those with NRG1 gene fusions or high HER3 expression, HMBD-001 aims to inhibit cancer cell growth and survival[4][5]. The drug exerts its effects through multiple mechanisms including antagonism of the ERBB-3 receptor (HER3), induction of antibody-dependent cell cytotoxicity (ADCC), and stimulation of T lymphocytes[3]. Clinical trials have shown that it is safe and well-tolerated as monotherapy up to high doses, with ongoing studies evaluating its efficacy alone or in combination with other agents such as enzalutamide for metastatic castration-resistant prostate cancer[8].
