HMBD-001 + cetuximab is an investigational combination therapy of two monoclonal antibodies: **HMBD-001**, a novel anti-HER3 (ERBB3) monoclonal antibody developed by Hummingbird Bioscience, and **cetuximab**, a clinically approved anti-epidermal growth factor receptor (EGFR, also known as ERBB1) monoclonal antibody. HMBD-001 blocks HER3 signaling, targeting both ligand-dependent and ligand-independent activation by binding the HER3 dimerization interface and fully inhibiting downstream oncogenic signaling pathways such as MAPK and PI3K, which are implicated in tumor growth and resistance. Cetuximab binds to EGFR, preventing ligand-induced activation and receptor dimerization, thus inhibiting EGFR-mediated proliferation and survival signaling. This combination targets both HER3 and EGFR pathways, which are often co-expressed and implicated in resistance mechanisms in solid tumors. It is under investigation for advanced HER3-positive or EGFR-positive solid tumors and tumors harboring NRG1 fusions, primarily as part of basket trials and in specific solid tumor indications, including non-small cell lung cancer and pancreatic cancer. The combination is at the early clinical development stage, and most published clinical results as of mid-2024 pertain to HMBD-001 as monotherapy or in combination with agents other than cetuximab[1][2][3][4][5].
