HMBD-002 is a non-depleting, high-affinity IgG4 monoclonal antibody targeting VISTA (V-domain Ig suppressor of T-cell activation), a negative immune checkpoint regulatory protein. It blocks VISTA's inhibitory function, reducing myeloid-derived suppression of T cell activity and preventing neutrophil migration. This stimulates a proinflammatory Th1/Th17 response and enhances antitumor immunity without depleting VISTA-expressing cells. Preclinical studies showed significant tumor growth inhibition in models of colorectal, breast, and lung cancer. HMBD-002 is being developed as both monotherapy and in combination with pembrolizumab for advanced solid tumors expressing VISTA, including triple-negative breast cancer (TNBC) and non-small cell lung cancer (NSCLC). It is currently in phase 1/2 clinical trials to evaluate safety, pharmacokinetics, immunological effects, and antitumor activity.
