**HMBD-002 + pembrolizumab** is a combination therapy consisting of HMBD-002, a humanized, non-depleting IgG4 monoclonal antibody targeting VISTA (V-domain Ig suppressor of T cell activation), and pembrolizumab, a PD-1 inhibitor marketed as KEYTRUDA. HMBD-002 acts by binding to a specific site on VISTA, blocking its immunosuppressive effects in the tumor microenvironment and enhancing anti-tumor immunity without depleting VISTA-expressing cells. Pembrolizumab is a programmed death-1 (PD-1) immune checkpoint inhibitor that blocks PD-1 to restore T cell activity against cancer. Preclinical models show that the combination leads to synergistic anti-tumor effects, and it is being evaluated for various solid tumors, with primary indications including triple-negative breast cancer (TNBC) and non-small cell lung cancer (NSCLC)[1][2][3][5][7][9].
