HMBD-011 is a potentially first-in-class, fully human monoclonal antibody that selectively targets the VH4-34 immunoglobulin heavy chain variable region. VH4-34+ B cells and autoantibodies are implicated in the pathogenesis of several autoimmune diseases, including systemic lupus erythematosus (SLE), cutaneous lupus erythematosus (CLE), and cold agglutinin disease (CAD). Developed using Hummingbird Bioscience’s Rational Antibody Discovery platform, HMBD-011 binds to a small conserved conformational hydrophobic patch within the FR1 region of VH4-34. This enables highly selective binding to pathogenic VH4-34+ antibodies and autoreactive B cells without off-target effects on other B cell populations. The mechanism involves direct clearance of circulating VH4-34+ antibodies as well as depletion of autoreactive B cells via antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Preclinical studies have demonstrated rapid elimination of pathogenic antibodies and effective inhibition of cold agglutination, with preservation of normal immune function due to its selectivity[1][3][5][7].
