HMBD-501 is a next-generation antibody-drug conjugate (ADC) targeting the HER3 (ERBB3) receptor. Developed using Hummingbird Bioscience’s proprietary antibody discovery and engineering platform, it features an Fc-engineered anti-HER3 monoclonal antibody conjugated to exatecan, a DNA topoisomerase I inhibitor payload. The ADC binds a novel epitope on HER3 that allows for effective targeting regardless of NRG1 ligand levels, potentially enabling activity in tumors with high ligand expression. Preclinical studies have demonstrated robust efficacy and tolerability in solid tumor models. Endeavor BioMedicines has acquired exclusive worldwide rights to develop and commercialize HMBD-501[1][2][3][4][7].
