Overview

HMBD-802 is a novel, first-in-class anti-HER2 dual-payload antibody-drug conjugate (ADC) developed by Hummingbird Bioscience. Built using the company's proprietary antibody and dual-payload ADC platform technologies, the agent is designed to deliver two distinct cytotoxic payloads directly to HER2-expressing tumor cells. This dual-payload approach aims to maximize anti-tumor efficacy and overcome resistance mechanisms associated with single-payload ADCs, specifically topoisomerase 1 inhibitor resistance (such as that seen with trastuzumab deruxtecan). By optimizing targeted delivery, HMBD-802 is intended to enhance the therapeutic index by reducing systemic toxicity while maintaining potent activity in HER2-positive and T-DXd-insensitive cancer settings.
