HMGA1 shRNA is a research-stage experimental RNA interference (RNAi) construct designed to silence the expression of the High Mobility Group AT-hook 1 (HMGA1) gene. HMGA1 is a chromatin-associated architectural transcription factor that is frequently overexpressed in various human cancers, including pancreatic cancer, breast cancer, and myeloproliferative neoplasms (MPN), where it drives disease progression by rewiring transcriptional networks and inducing genes involved in cell cycle progression and cell fate (such as GATA2). Because HMGA1 lacks a traditional enzymatic pocket, it is difficult to target with small molecules, making shRNA a valuable tool for preclinical validation. In laboratory settings, HMGA1 shRNA is often delivered via viral vectors (lentivirus or adenovirus) to inhibit proliferation, induce apoptosis, and impair clonogenicity in cancer cell lines and animal models.
