HMGB1 A box is a recombinant protein fragment corresponding to the A-box domain of the High Mobility Group Box 1 (HMGB1) protein. HMGB1 is a damage-associated molecular pattern (DAMP) that triggers neuroinflammation by binding to receptors such as RAGE and TLR4. The A-box domain acts as a natural competitive antagonist to full-length HMGB1, effectively blocking its pro-inflammatory signaling. In preclinical models of Parkinson's disease, HMGB1 A box has demonstrated the ability to restrict microglia activation, up-regulate the CD200-CD200R signaling axis, and inhibit the differentiation and infiltration of Th17 cells. These actions help restore dopaminergic neurons and reduce inflammatory mediators like TNF-α and IL-1β. It is primarily utilized as a research tool to investigate HMGB1-mediated pathology and is being explored as a potential therapeutic candidate for neuroinflammatory and autoimmune disorders.
