**HMGN1-gp100 DNA** is an experimental plasmid DNA cancer vaccine encoding a fusion of the melanoma-associated antigen gp100 and high mobility group nucleosome-binding protein 1. Following cellular uptake and expression, the fusion protein is intended to promote gp100-specific antitumor immunity, while HMGN1 acts as an endogenous alarmin immunoadjuvant that enhances Th1-polarized cellular immune responses. The vaccine was evaluated in preclinical mouse melanoma models, including B16F1 challenge experiments, and has also been formulated in cationic self-assembling peptide hydrogels to improve local plasmid retention and immunostimulation. It is an academic National Cancer Institute research program with no identified commercial sponsor, clinical trial, marketed product, or assigned INN.
