HMI-115 is a first-in-class monoclonal antibody that targets and non-competitively blocks the prolactin receptor (PRLR). It was initially developed by Bayer and is now being developed by Hope Medicine, which holds global rights through an exclusive license agreement. The drug’s mechanism of action involves inhibiting PRLR-mediated signaling, a pathway implicated in diseases with dysregulated prolactin activity. HMI-115 is under clinical development for the treatment of endometriosis-associated pain and androgenetic alopecia (pattern hair loss), both conditions where prolactin signaling plays a pathogenic role. In Phase II trials for endometriosis, HMI-115 demonstrated significant reductions in dysmenorrhea and non-menstrual pelvic pain without inducing perimenopausal symptoms or affecting sex hormone levels. For androgenetic alopecia, early studies showed increased hair density in treated patients. The drug has been granted Breakthrough Therapy Designation by China’s NMPA for moderate to severe endometriosis-associated pain[2][3][5][8].
