hMN-14 × m734 is an investigational, chemically assembled F(ab')2 bispecific antibody construct for pretargeted radioimmunotherapy of carcinoembryonic antigen-expressing cancers. One Fab' arm is derived from the humanized anti-carcinoembryonic antigen antibody hMN-14, while the other is derived from murine anti-DTPA-indium antibody m734. The construct is administered first to localize at carcinoembryonic antigen-positive tumor sites; a separately administered iodine-131-labeled di-DTPA-indium bivalent hapten subsequently binds the m734 arm, concentrating therapeutic radioactivity at the tumor. Clinical studies evaluated this pretargeting system in nonmedullary thyroid carcinoma and metastatic medullary thyroid carcinoma, including a prospective phase II study in medullary thyroid carcinoma.
