HMP19 (also known as FAM171A2) is a type II, single-pass transmembrane protein that functions as a metastasis suppressor in pancreatic ductal adenocarcinoma (PDAC). Localized to the Golgi apparatus and involved in vesicular trafficking, HMP19 is secreted within extracellular vesicles (EVs). These HMP19-containing EVs have been shown to inhibit the migration of metastatic PDAC cells by interacting with and downregulating the Epidermal Growth Factor Receptor (EGFR) signaling pathway. This downregulation leads to reduced EGFR phosphorylation and protein expression, which subsequently attenuates nuclear ERK1/2 activity. The protein was identified through genome-wide shRNA screening and is being investigated for its potential as a therapeutic agent or biomarker in advanced pancreatic cancer.
