HMPL-011 is an orally bioavailable, synthetic small molecule cytokine modulator developed by Hutchison MediPharma (now HUTCHMED). It functions by inhibiting the activation of the NF-κB (nuclear factor kappa B) signaling pathway, which in turn suppresses the production of multiple pro-inflammatory cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6). The drug was primarily investigated for the treatment of inflammatory bowel diseases (IBD), such as Crohn's disease and ulcerative colitis. A Phase I clinical trial (NCT00986375) was conducted in healthy volunteers to assess its safety, tolerability, and pharmacokinetics. However, development of HMPL-011 appears to have been discontinued, as it is no longer listed in the company's active development pipeline.
