HMPL-453 + toripalimab is a combination investigational therapy composed of two distinct agents. HMPL-453 is a novel, highly selective, and potent small molecule inhibitor of fibroblast growth factor receptors (FGFR) 1, 2, and 3, designed to block aberrant FGFR signaling that drives tumor growth, angiogenesis, and treatment resistance, primarily in solid tumors such as intrahepatic cholangiocarcinoma (IHCC)[3][4]. Toripalimab is a recombinant humanized monoclonal antibody that targets programmed death receptor-1 (PD-1), functioning as an immune checkpoint inhibitor that reinvigorates T-cell responses against malignancies, and is approved or in clinical trials for various cancers including non-small-cell lung cancer (NSCLC) and nasopharyngeal cancer[8][6]. The combination is intended to harness targeted FGFR blockade alongside immune checkpoint inhibition, potentially offering synergistic anti-tumor activity.
