Overview

HMPL-653 is an investigational, orally bioavailable, highly selective, and potent small molecule inhibitor of the macrophage colony-stimulating factor 1 receptor (CSF-1R). It is being developed by HUTCHMED for the treatment of advanced or metastatic solid tumors and tenosynovial giant cell tumor (TGCT). CSF-1R is typically expressed on macrophages and plays a role in their growth and differentiation. By inhibiting CSF-1R signaling, HMPL-653 aims to modulate the tumor microenvironment, relieve tumor immunosuppression, and potentially synergize with other anti-cancer therapies such as immune checkpoint inhibitors. The drug is currently in Phase I clinical trials in China[2][3][5][7].
