HMPL-A83 is an investigational humanized IgG4 monoclonal antibody that targets the cell surface antigen CD47. Developed by HUTCHMED, it functions as an immune checkpoint inhibitor by blocking the interaction between CD47 and signal regulatory protein alpha (SIRPα), thereby disrupting the "do not eat me" signal that cancer cells use to evade macrophage-mediated phagocytosis. This mechanism enhances the immune system's ability to recognize and destroy tumor cells. Preclinical studies have shown that HMPL-A83 has high affinity for CD47 on tumor cells, induces strong phagocytosis of multiple tumor types, demonstrates low affinity for red blood cells, and does not induce hemagglutination—suggesting a lower risk of anemia compared to other anti-CD47 agents. It is currently being evaluated in Phase I clinical trials for advanced malignant neoplasms[1][2][3][4][5][6][7].
