HMR4011 is a small molecule inhibitor of cholesteryl ester transfer protein (CETP) that was developed by Aventis (now Sanofi) for the treatment of dyslipidemia and atherosclerosis. CETP is a plasma protein that facilitates the transport of cholesteryl esters and triglycerides between lipoproteins, specifically moving cholesteryl esters from high-density lipoprotein (HDL) to very-low-density lipoprotein (VLDL) and low-density lipoprotein (LDL). By inhibiting this process, HMR4011 aims to increase HDL cholesterol levels and decrease LDL cholesterol levels, thereby potentially reducing the risk of cardiovascular disease. Development of HMR4011 reached Phase I clinical trials but was discontinued in the early 2000s, reflecting the broader challenges and safety concerns encountered by the CETP inhibitor class during that period.
