Overview

hN1 mAb is a humanized monoclonal antibody that selectively targets the negative regulatory region (NRR) of the Notch1 receptor. Developed by Pfizer, this antibody is designed to inhibit Notch1 signaling by binding to the NRR and preventing the conformational changes necessary for proteolytic activation by ADAM proteases and the gamma-secretase complex. Notch1 signaling is a critical pathway in the maintenance and self-renewal of leukemia progenitor cells, particularly in T-cell acute lymphoblastic leukemia (T-ALL), where activating mutations in Notch1 are present in approximately 50% of cases. Preclinical research has shown that hN1 mAb effectively reduces leukemic engraftment and induces apoptosis in Notch1-mutated T-ALL models, suggesting potential therapeutic utility in targeting leukemia stem cell populations.
