hnCD16-iNK cells are an investigational allogeneic natural killer (NK) cell therapy engineered to express a high-affinity, non-cleavable human CD16 (FcγRIIIA) receptor, designed to enhance antibody-dependent cellular cytotoxicity (ADCC) against tumor cells when combined with therapeutic monoclonal antibodies. By stably augmenting surface CD16, these induced NK (iNK) cells aim to provide potent off-the-shelf cytotoxic activity with improved engagement of IgG1 antibody Fc domains, leveraging NK-cell–mediated killing while minimizing risks of graft-versus-host disease and severe cytokine release syndrome commonly associated with T cell therapies.[3][5]
