HNF4α ASO refers to an Antisense Oligonucleotide (ASO) designed to target Hepatocyte Nuclear Factor 4 alpha (HNF4α). HNF4α is a nuclear receptor and a master regulator of liver development and function, playing a critical role in metabolic homeostasis, including glucose and lipid metabolism, and the development of organs like the liver, kidney, and intestines. Its expression is often down-regulated in chronic and inflammatory liver diseases, such as cirrhosis and non-alcoholic fatty liver disease (NAFLD), and mutations in the HNF4A gene are linked to Maturity Onset Diabetes of the Young 1 (MODY1) and liver cancer. An ASO targeting HNF4α would typically function by binding to the HNF4α messenger RNA (mRNA), thereby modulating its expression. This modulation can occur through several mechanisms, including RNase H-dependent mRNA degradation, alteration of pre-mRNA splicing, or steric hindrance to inhibit translation. Such an approach aims to therapeutically intervene in diseases where aberrant HNF4α expression or function is implicated, potentially by reducing or increasing HNF4α protein levels or altering its isoforms.
