HO-4073 is a synthetic small molecule curcumin analog belonging to the diarylidenylpiperidone (DAP) class. Developed by researchers at Ohio State University and the University of Pécs, it was designed as a parafluorinated variant to improve upon the poor pharmacokinetic properties of natural curcumin. HO-4073 functions as a potent inhibitor of the JAK/STAT3 signaling pathway, which is often constitutively active in various malignancies. Preclinical research has demonstrated its efficacy in inhibiting cell proliferation and inducing apoptosis in head and neck squamous cell carcinoma (HNSCC) cell lines. Notably, HO-4073 has shown the potential to sensitize resistant cancer cells to cisplatin-based chemotherapy and radiotherapy, suggesting a role in overcoming treatment resistance in HNSCC.
