Overview

Holmium-166-DOTMP is an experimental radiopharmaceutical developed for skeletal targeted radiotherapy (STR), primarily in combination with high-dose chemotherapy. It consists of the chelating agent DOTMP, which localizes to bone, and the radioactive isotope holmium-166, a beta-emitting radionuclide. When administered, the compound accumulates in bone tissue and delivers localized ionizing radiation to tumor sites within the bone and bone marrow. This mechanism is intended to destroy cancer cells—particularly myeloma cells—in these locations. The drug was developed by NeoRx Corporation (originator Dow Chemical) as a potential treatment for multiple myeloma and other cancers with skeletal involvement. Clinical development reached phase 3 trials for multiple myeloma but was discontinued before approval[1][2][3][4].
