Overview

Homogeneous anti-EGFR ADC 1 is an experimental antibody-drug conjugate (ADC) designed for the treatment of glioblastoma multiforme (GBM). It consists of the anti-EGFR monoclonal antibody cetuximab site-specifically conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) using the AbClick technology. This specific construct is characterized by its high homogeneity and a defined drug-to-antibody ratio (DAR) of 2. Research indicates that these parameters are critical for overcoming the blood-brain barrier (BBB), as lower-DAR homogeneous species demonstrate superior brain tumor penetration and therapeutic efficacy compared to heterogeneous or highly loaded ADCs. In preclinical studies, it has shown significant survival benefits in orthotopic brain tumor models by optimizing payload delivery to intracranial tumors.
